Blocking a pathway at two points is the textbook recipe for a synergy, and that is the whole theoretical case.

The 1999 trial is where it was tested. A green tea extract delivering 270 mg of EGCG a day alongside 150 mg of caffeine raised 24-hour energy expenditure by about four per cent and shifted the respiratory quotient toward fat oxidation. The same experiment ran caffeine alone at 150 mg as a separate arm, and that arm moved nothing.

Pooling the energy-expenditure trials that followed put the combined effect at a level best described as measurable rather than large, and the review of the interaction notes that habitual caffeine intake blunts it: the people in whom catechin and caffeine mixtures work best are the ones who drink the least coffee.

Why the amount decides this one

Hydroxycitric acid and fat synthesis

HCA inhibits ATP-citrate lyase, an enzyme in the pathway that builds fat from surplus carbohydrate. The biochemistry is settled and has been since the nineteen-seventies. The question was always whether inhibiting it changes anything in a person eating an ordinary diet, and the pooled trial evidence is where that question was answered rather than in the enzymology.

Raspberry ketone and lipolysis in culture

In cultured fat cells raspberry ketone increases lipolysis and adiponectin secretion, which is the entire basis for its presence on labels like this one. The cleanest animal test fed it to mice on a high-fat diet and reported it did not reduce adiposity beyond the effect of the animals eating less. “The ingredients must add up to something.”
